Study on anti-angiogenesis effect of curcumin pigments and expression of their relevant factors

 

In order to get different function of anti-angiogenesis on the effect of three curcumin pigments ( curcumin, demethoxycurcumin, bisdemethoxycurcumin ), the researching mainly testing the inhibition function of the The OX – LDL induced human umbilical vein endothelial cells HUVEC proliferation on the effect of curcumin pigments and the influence on HUVEC cell migration.  Using the method of chick chorioallantoic membrane (CAM) to see the effect of curcumin on angiogenesis; RT-PCR testing the VEGF expression of HUVEC cell growth factor ;RT-PCR testing the influence of Cell adhesion factor ICAM-1, VCAM-1 from HUVEC.

 

From these testing found that the effect of curcumin pigments which in the measuring range of 4,8,16 mg·L-1, they all act the inhibiting effect to HUVEC cell proliferation which be induced by OX-LDL and the inhibition effect relationship on dose-response.And the proliferation  effective of curcumin is stronger than the other two derivatives (P<0.01).  All of these three effect of curcumin pigments can inhibit of  the HUVEC  cell migration, inhibit the angiogenesis of the chick embryo chorioallantoic membrane (CAM). The migration inhibition ratio of high purity of curcumin is greater than the demethoxycurcumin, bisdemethoxycurcumin. (P<0.01). All of these three curcumin can lower the expression of VEGF, ICAM-1.  For the lower function to VEGF of curcumin is greater than the demethoxycurcumin and bisdemethoxycurcumin( P< 0.01). For the lowering effection of ICAM-1, the bisdemethoxycurcumin showed the greater function than others (P<0.01). All of the three curcumin have little effection on the expression of VCAM-1 and only the bisdemethoxycurcumin have the lowering function effective ( P<0.05).

Through these testing study, we can see all of these three curcumin pigments have the effective function of resisting angiogenesis, inhibiting proliferation and migration of endothelial cells, down-regulating the expression of VEGF and adhesion molecules ICAM-1.
